Pachypodostyflavone, a New 3-methoxy Flavone and Other Constituents with Antifilarial Activities from the Stem Bark of Duguetia staudtii

Abstract A new flavone derivative named pachypodostyflavone (1), along with 8 known compounds (2–9) and a mixture of β-sitosterol stigmasterol were isolated from the stem bark Duguetia staudtii (Annonaceae), based on bioassay-guided fractionation. Their structures determined using high-resolution mass spectrometry NMR spectroscopic data, as well by comparison literature values their analogs. Selected evaluated for in vitro antifilaricidal activities Onchocerca ochengi microfilariae adult worms. Inhibition motility was spectroscopically microfilaria male Viability female worms MTT/ Formazan assay. Auranofin at 10 µM 2% DMSO used positive negative controls, respectively. Compounds 1 7 showed potent anti-onchocerca 100% activity 250 µg/mL both O. worms, while compound 5 displayed 30 µg/mL.
